Jafferi was shot dead by gunmen outside his home in Peshawar at 8 a.m., when he was going to office in his car, according to police.
The murder of a consulate employee represents a new tactic by extremists hoping to isolate Pakistan from its supporters in the international community, Jeremiah Knight, the Acting Spokesman of the US embassy said in a statement.
"It will not succeed," the Islamic republic news agency quoted him as saying.
"The international community is firm in its support of the Pakistani people and their democratic government," the spokesman said.
